In a simple workflow, the sample is combined with a reagent cocktail that includes cell wall degrading enzymes and the thermophilic protease. Following cell wall digestion, the sample is heated to 75°C to lyse the cells, inactivate nucleases, and activate the thermostable protease. Then, the sample is heated to 95°C to inactivate the protease.
